Innovative Approaches for the Identification and Development of Cancer Fighting Small Molecules

Cancer is currently one of the most important global public health concerns, serving as the leading cause of death for people globally. Cancer diagnoses have increased globally in recent years to unprecedented proportions, accounting for over 10 million deaths per year. There are about 100 different forms of cancer, including colorectal, lung, breast, ovarian, prostate, and pancreatic cancers. As a result, the anticancer therapeutic arsenal should be continually supplemented with novel small compounds that preferentially target tumour cells while causing minimum toxicity to normal cells.

Although several anticancer drugs have been approved in recent years, the discovery of new and effective drug candidates has been hampered with the increased attrition rate caused by the limited therapeutic windows, drug resistance, and inadequate efficacy. Accordingly, there exists a significant imperative to identify and exploit promising drug leads from the perspective of drug design and medicinal chemistry, in order to facilitate the development of more effective therapeutic interventions for various types of human cancers.

To this end, specialists in all fields of drug discovery and medicinal chemistry are invited to contribute to this Article Collection, highlighting cutting-edge approaches and reporting their most recent accomplishments in drug design and development of innovative anticancer prospects. The goal of the Article Collection is to compile original research papers as well as reviews that discuss the most current breakthroughs in the identification of synthetic and natural anticancer molecules, endowed with effective enzyme inhibition/interactions, that exhibit intriguing potential as therapeutic leads.

The manuscripts of interest (Original articles and Reviews articles) may include, but are not limited to:

  • Synthesis and semi-synthesis of new anticancer molecules.
  • Identification and isolation of anticancer agents from natural sources.
  • Drugs-repurposing opportunities for cancer therapy.
  • Exploiting the Computer-Aided Drug Design approaches to develop innovative anticancer agents, supported by experimental validation.
  • Drug targeting strategies in cancer treatment.
